Application

Anti-phospho-mTOR (pSer2481) antibody produced in rabbit has been used in western blot analysis.

Biochem/physiol Actions

Mechanistic target of rapamycin kinase (MTOR) recognizes various intracellular and extracellular signals and processes them to regulate cell growth, metabolism, proliferation and survival. mTOR is also involved in mitochondrial respiration by activating oxidative phosphorylation. Deregulation of mTOR phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) complex signalling pathway, leads to leukaemogenesis. Mutations leading to aberrant expression of mTOR causes cancer cachexia. mTOR plays a key role in synaptic plasticity and autophagy. Polymorphism of mTOR is associated with brain malformation and various neuropsychiatric disorders like brain ischemia, autism, epilepsy, schizophrenia etc. mTOR is a key regulator of anabolic and catabolic processes. mTOR is activated by insulin or insulin like growth factor 1 (IGF1) and regulate PI3K/Akt/mTOR/tuberous sclerosis (TSC)/the small GTPase (guanidine triphosphatase) Ras (rat sarcoma) homologue enriched in brain (RHEB) signalling pathway. mTOR is a key regulator of lipid biogenesis by influencing lipogenic genes like sterol regulatory element-binding protein 1 (SREBP1). Dysregulation of mTOR is associated with type 2 diabetes and obesity.

General description

Mechanistic target of rapamycin kinase (MTOR) is a serine/threonine kinase. mTOR belongs to the family of phosphatidylinositol kinase-related kinase (PIKK). mTOR exists as two different complexes, mTOR complex 1 (mTORC1) and mTOR complex 2 (mTORC2). In human chromosome, the gene MTOR is localized on 1p36.22.

Immunogen

Peptide sequence around phosphorylation site of Serine 2481(I-H-S(p)-F-I) derived from Human mTOR.

Physical form

Supplied at 1.0mg/mL in phosphate buffered saline (without Mg2+ and Ca2+), pH 7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol.

Specificity

The antibody detects endogenous level of mTOR only when phosphorylated at Serine 2481.
